Mechanism of Action:
Rabeprazole functions as an irreversible inhibitor of the hydrogen-potassium adenosine triphosphatase (H+/K+-ATPase) enzyme system, commonly known as the proton pump, found in the secretory surface of parietal cells in the stomach. By blocking this final step of acid production, it effectively reduces the secretion of both basal and stimulated gastric acid, regardless of the stimulus.
Indications:
Rabeprazole sodium is primarily indicated for conditions caused by excessive or problematic stomach acid production:

• Gastroesophageal Reflux Disease (GERD): Healing and maintenance of healing of erosive or ulcerative GERD in adults and adolescents (12 years and older).
• Treatment of symptomatic, non-erosive GERD.
• Peptic Ulcer Disease: Healing of duodenal ulcers and gastric ulcers.
• Helicobacter pylori (H. pylori) Eradication: Used in combination with specific antibiotics (e.g., amoxicillin, clarithromycin) as a triple-therapy regimen to treat H. pylori infection in patients with peptic ulcer disease, reducing the risk of recurrence.
• Zollinger-Ellison Syndrome: Long-term management of hypersecretory conditions, including Zollinger-Ellison syndrome (a rare condition causing severe acid overproduction due to a tumor).

Critical Safety Information:
Administration: Best taken 30 minutes before a meal (typically breakfast) for optimal acid control.Tablets should be swallowed whole and not chewed, crushed, or split.
Duration of Use: Use for longer than recommended treatment periods may increase certain risks.
Associated Risks (Long-Term Use):
Bone Fractures: Increased risk of hip, wrist, and spine fractures with prolonged use (typically a year or longer).
Infections: Slightly increased risk of certain infections, including Clostridioides difficile (C. diff) associated diarrhea.
Vitamin B12 Deficiency: Long-term use can reduce the absorption of Vitamin B12.
Hypomagnesemia: May cause low magnesium levels in the blood.
